COPENHAGEN, Denmark--(BUSINESS WIRE)--Dec 8, 2024-- Genmab A/S (Nasdaq: GMAB): Genmab A/S (Nasdaq: GMAB) today announced results from the Phase 1b/2 EPCORE ® CLL-1 clinical trial evaluating epcoritamab (Abstract #883), a T-cell engaging bispecific antibody administered subcutaneously, demonstrated an overall response rate (ORR) of 61 percent and a complete response (CR) rate of 39 percent in difficult-to-treat adult patients with relapsed or refractory (R/R) chronic lymphocytic leukemia (CLL) treated with epcoritamab monotherapy. These results, from the monotherapy expansion (EXP) cohort (n=23) of the trial, along with the first safety data from the optimization (OPT) cohort, were presented at the 66 th Annual Meeting and Exposition of the American Society of Hematology (ASH), during the ASH Annual Meeting Press Program. The data will be presented during an oral session on December 9, 2024. In the EXP cohort, the median time to response was two (2.0) months and the median time to CR was 5.6 months. Among all patients in the cohort, median progression-free survival (PFS) was 12.8 months and median overall survival (OS) was not reached (median follow-up was 22.8 months). An estimated 65 percent of patients were alive at 15 months. Among 12 responders evaluable for minimal residual disease (MRD) by next-generation sequencing in peripheral blood, nine patients (75 percent) had undetectable MRD. The most frequent non-hematologic treatment-emergent adverse events (TEAEs) in the EXP cohort were cytokine release syndrome (CRS; 96 percent), diarrhea (48 percent), peripheral edema (48 percent), fatigue (43 percent), and injection-site reaction (43 percent). Cytopenias were common (anemia, 65 percent; thrombocytopenia, 65 percent; neutropenia, 48 percent); however, most patients had baseline anemia and thrombocytopenia, suggesting that these events were largely disease-related. Three cases of immune effector cell-associated neurotoxicity syndrome (ICANS) were reported (one Grade 1; two Grade 2), and there was one clinical tumor lysis syndrome (CTLS) case (Grade 2). These cases did not lead to treatment discontinuation. Four fatal TEAEs occurred - two cases of pneumonia, one case of sepsis and one case of squamous cell carcinoma of the skin. The EXP cohort followed a 2-step step-up dose regimen, and CRS was manageable and primarily low grade (9 percent Grade 1, 70 percent Grade 2, 17 percent Grade 3). In the first data from the separate OPT cohort, which followed a 3-step step-up dose regimen, CRS severity was substantially reduced with only low-grade events (71 percent Grade 1, 12 percent Grade 2). In both cohorts, CRS events primarily occurred following the first full dose, and none led to treatment discontinuation. No ICANS or CTLS cases were reported in the OPT cohort. “These EPCORE CLL-1 data are encouraging, especially as the majority of patients were heavily pre-treated with at least four lines of therapy,” said Alexey Danilov, MD, PhD, Marianne and Gerhard Pinkus, Professor and Director of Early Clinical Therapeutics and Associate Director of the Toni Stephenson Lymphoma Center, Department of Hematology and Hematopoietic Cell Transplantation, City of Hope. “Despite progress in treating chronic lymphocytic leukemia, there remains a tremendous need for additional therapeutic options for high-risk patients whose disease has progressed following standard chemoimmunotherapy and targeted therapies.” Additional data from the EXP cohort showed high response rates in patients with high-risk factors treated with epcoritamab, including TP53 aberrations, IGHV-unmutated disease and double-exposed disease – prognostic markers that are associated with disease progression and decreased survival. i,ii,iii In patients with TP53 aberrations (n=15), the ORR was 67 percent with a CR of 33 percent. Among patients with IGHV-unmutated disease (n=16), the ORR was 63 percent, and the CR was 44 percent. In double-refractory patients, the ORR was 53 percent, and the CR was 37 percent. All patients in the trial had prior chemoimmunotherapy, and most patients had previously received targeted therapies such as BTK and BCL2 inhibitors (double-exposed) and had high-risk disease characteristics. “Chronic lymphocytic leukemia is incurable, and patients often need a variety of treatments throughout their lifetime, especially if their disease has high-risk prognostic factors, has relapsed or has become refractory to the current standard-of-care, including targeted therapies,” said Dr. Judith Klimovsky, Executive Vice President & Chief Development Officer, Genmab. “These early data show the potential therapeutic applicability of epcoritamab in relapsed or refractory chronic lymphocytic leukemia, and further reinforce the potential of epcoritamab as a core therapy for the treatment of B-cell malignancies.” Use of epcoritamab in CLL is not approved in the U.S. or in the EU or in any other territory. NEW YORK: Investors are expecting more gains for the US stock market in 2025 after two straight standout years, fueled by a solid economy supporting corporate profits, moderating interest rates and pro-growth policies from incoming president Donald Trump. The benchmark S&P 500 up over 23 percent year-to-date, even with a recent speed bump, and is on pace for its second straight year of gains exceeding 20 percent, lifted by megacap tech stocks and excitement over the business potential of artificial intelligence. Investors are more confident about the economy than this time a year ago, with consumers and businesses having absorbed higher interest rates and the Federal Reserve now lowering them - albeit by not as much as hoped. Corporate profits are also expected to be strong, with S&P 500 earnings projected to rise 14 percent in 2025, according to LSEG IBES. On the other side of the ledger, inflation remains stubborn, and Wall Street is wary of a rebound that could lead the Fed to change course on its easing cycle. Indeed, stocks pulled back sharply on Wednesday after the central bank projected fewer rate cuts next year as it braced for firmer inflation. Such prospects could become more likely if Trump implements tariffs on US imports that lead to higher consumer prices. Stock valuations, meanwhile, are around their steepest levels in more than three years, leaving greater potential for turbulence. “We’ve been on quite the tear coming off the lows back at the end of 2022. It’s been pretty eye-watering,” said Garrett Melson, portfolio strategist at Natixis Investment Managers. “Animal spirits... are certainly running pretty wild right now, but you might need to temper that a little bit as you start to move through the year,” said Melson, who thinks the stock market could still produce solid gains of around 10 percent in 2025 if not the returns of the prior two years. Wall Street firms are mostly projecting gains for the market next year, with S&P 500 year-end targets ranging from 6,000 to 7,000. The index was last hovering around 5,900. Optimistic investors can point to a bull market that is neither old nor over-extended, by historic measures. The current bull market for the S&P 500 that began in October 2022 is less than half as long as the average length of the 10 prior ones, according to Keith Lerner, co-chief investment officer at Truist Advisory Services. The S&P 500’s roughly 64 percent gain during this latest run trails the 108 percent median gain and 184 percent average rise of the prior bull markets, according to Lerner. “If you zoom out a little bit, yes, we have a lot of gains, but if you look at a typical bull market, it suggests that we still have further gains to go,” Lerner said. Other historic signs also bode well. The S&P 500 has gained an average of 12.3 percent following the eight instances of back-to-back 20 percent annual gains since 1950, according to Ryan Detrick, chief market strategist at Carson Group, compared to a 9.3 percent overall average increase over that time. The index increased six of the eight times. Bolstering the upbeat sentiment is the prevailing sense on Wall Street that the economy has weathered the rate hikes the Fed implemented starting in 2022 to quell inflation. A Natixis Investment Managers survey conducted in recent weeks found 73 percent of institutional investors said the US will avoid a recession in 2025. That’s a sharp turnaround from a year ago, when 62 percent projected such a downturn in the coming year. Citigroup’s economic surprise index, which measures how economic data performs versus expectations, has been solidly positive for the past two months, another rosy sign for investors. Adding to expectations of a solid economy, Trump is expected to pursue an agenda that includes tax cuts and deregulation that supports growth. “We’re leaving 2024 on pretty good footing, and we think there is some re-acceleration in 2025,” said Sameer Samana, senior global market strategist at Wells Fargo Investment Institute. “Markets tend to front-run the economy, so they will position for that economic re-acceleration sooner rather than later.” However, stocks are also leaving 2024 at elevated valuations: The S&P 500 is trading at nearly 22 times expected earnings over the next 12 months, according to LSEG. That is well above its long-term average of 15.8, and not far from the 22.6 level it reached earlier this month, its highest since early 2021. Investors maintain that valuations can stay high for long periods and do not necessarily indicate imminent declines. But future gains may rest more on earnings growth, while higher valuations could make stocks more easily rattled by any disappointments. Risks include policy uncertainty such as Trump’s expected push to raise tariffs on imports from China and other trading partners, which analysts estimate could hurt corporate profits. Higher tariffs could also increase inflation, which is another worry for investors. The pace of inflation has fallen dramatically since hitting 40-year highs in 2022, but remains above the Fed’s 2 percent target. Ghana's former president John Drahami Mahama won a historic comeback election victory on Sunday after the ruling party accepted defeat with voters appearing to punish them for the government's handling of an economic crisis. Following Saturday's election, New Patriotic Party candidate Vice President Mahamudu Bawumia on Sunday conceded Ghanaians wanted change after he failed to shake off widespread frustration over high costs of living. His defeat ended eight years in power for the NPP under President Nana Akufo-Addo, whose last term was marked by the west African state's worst economic turmoil in years, high inflation and a debt default. For opposition National Democratic Congress party's Mahama, president from 2012-2017, it was his third attempt to reclaim the top post after falling short in 2016 and 2020 elections. "Former president Mahama has won the presidential election decisively," Bawumia told a press conference. Mahama's NDC also won the parliament elections, Bawumia said, referring to the NPP's own internal collation of election results. "The people of Ghana have spoken, the people have voted for change at this time and we respect it with all humility," he said. The speedy concession came as official vote tallies were still arriving. Electoral authorities have yet to announce the results for the presidential and parliament ballots. Blaring horns and whistles and waving the party's green, white and red flags, Mahama supporters took the streets to celebrate outside the party campaign office in the capital Accra. "They said he can't come back, and he has come. The nation builder is back to build our Ghana for us," said trader and Mahama supporter Leyla Alhassan enjoying the celebrations. Mahama has yet to speak publicly. But on his X account, Mahama confirmed he received Bawumia's congratulatory call over his "emphatic victory". The US embassy in Accra also applauded a "successful election that reflects the will of the Ghanaian people." Ghana's economic woes dominated the election, after the continent's top gold producer and world's second cacao exporter went through a crisis of default and currency devaluation, ending with a $3 billion IMF bailout. Earlier, NDC spokesman Sammy Gyamfi told reporters the party's internal review of results showed Mahama won 56.3 percent of the vote against 41.3 percent for Bawumia. Political parties had agents at polling stations to observe and tally the initial vote counts before the ballots were sent for official collation by the election commission. The commission had said official results were likely due by Tuesday. With a history of democratic stability, Ghana's two main parties, the NPP and NDC, have alternated in power equally since the return to multi-party politics in 1992. Under the slogan "Break the 8" -- a reference to two terms in power -- Bawumia had sought to lead the NPP to an unprecedented third term. But he struggled to break away from criticism of Akufo-Addo's economic record. Though inflation slowed from more than 50 percent to around 23 percent, and other macro-economic indicators are stabilising, economic struggles were still a clear election issue for many. Jimmy Carter, the United States’ longest-lived president, was never afraid of speaking his mind. Forthright and fearless, the Nobel Prize winner took pot-shots at former prime minister Tony Blair and ex-US president George W Bush among others. His death came after repeated bouts of illness in which images of the increasingly frail former president failed to erase memories of his fierce spirit. Democrat James Earl “Jimmy” Carter Jr swept to power in 1977 with his Trust Me campaign helping to beat Republican president Gerald Ford. Serving as 39th US president from 1977 to 1981, he sought to make government “competent and compassionate” but was ousted by the unstoppable Hollywood appeal of a certain Ronald Reagan. A skilled sportsman, Mr Carter left his home of Plains, Georgia, to join the US Navy, returning later to run his family’s peanut business. A stint in the Georgia senate lit the touchpaper on his political career and he rose to the top of the Democratic movement. But he will also be remembered for a bizarre encounter with a deeply disgruntled opponent. The president was enjoying a relaxing fishing trip near his home town in 1979 when his craft was attacked by a furious swamp rabbit which reportedly swam up to the boat hissing wildly. The press had a field day, with one paper bearing the headline President Attacked By Rabbit. Away from encounters with belligerent bunnies, Mr Carter’s willingness to address politically uncomfortable topics did not diminish with age. He recently said that he would be willing to travel to North Korea for peace talks on behalf of US President Donald Trump. He also famously mounted a ferocious and personal attack on Tony Blair over the Iraq war, weeks before the prime minister left office in June 2007. Mr Carter, who had already denounced George W Bush’s presidency as “the worst in history”, used an interview on BBC radio to condemn Mr Blair for his tight relations with Mr Bush, particularly concerning the Iraq War. Asked how he would characterise Mr Blair’s relationship with Mr Bush, Mr Carter replied: “Abominable. Loyal, blind, apparently subservient. “I think that the almost undeviating support by Great Britain for the ill-advised policies of President Bush in Iraq have been a major tragedy for the world.” Mr Carter was also voluble over the Rhodesia crisis, which was about to end during his presidency. His support for Robert Mugabe at the time generated widespread criticism. He was said to have ignored the warnings of many prominent Zimbabweans, black and white, about what sort of leader Mugabe would be. This was seen by Mr Carter’s critics as “deserving a prominent place among the outrages of the Carter years”. Mr Carter has since said he and his administration had spent more effort and worry on Rhodesia than on the Middle East. He admitted he had supported two revolutionaries in Mugabe and Joshua Nkomo, and with hindsight said later that Mugabe had been “a good leader gone bad”, having at first been “a very enlightened president”. One US commentator wrote: “History will not look kindly on those in the West who insisted on bringing the avowed Marxist Mugabe into the government. “In particular, the Jimmy Carter foreign policy... bears some responsibility for the fate of a small African country with scant connection to American national interests.” In recent years Mr Carter developed a reputation as an international peace negotiator. He won the Nobel Peace Prize in 2002 for his commitment to finding peaceful solutions to international conflicts, his work with human rights and democracy initiatives, and his promotion of economic and social programmes. Mr Carter was dispatched to North Korea in August 2008 to secure the release of US citizen Aijalon Mahli Gomes, who had been sentenced to eight years of hard labour after being found guilty of illegally entering North Korea. He successfully secured the release of Mr Gomes. In 2010 he returned to the White House to greet President Barack Obama and discuss international affairs amid rising tensions on the Korean peninsula. Proving politics runs in the family, in 2013 his grandson Jason, a state senator, announced his bid to become governor in Georgia, where his famous grandfather governed before becoming president. He eventually lost to incumbent Republican Nathan Deal. Fears that Mr Carter’s health was deteriorating were sparked in 2015 when he cut short an election observation visit in Guyana because he was “not feeling well”. It would have been Mr Carter’s 39th trip to personally observe an international election. Three months later, on August 12, he revealed he had cancer which had been diagnosed after he underwent surgery to remove a small mass in his liver. Mr Obama was among the well-wishers hoping for Mr Carter’s full recovery after it was confirmed the cancer had spread widely. Melanoma had been found in his brain and liver, and Mr Carter underwent immunotherapy and radiation therapy, before announcing in March the following year that he no longer needed any treatment. In 2017, Mr Carter was taken to hospital as a precaution, after he became dehydrated at a home-building project in Canada. He was admitted to hospital on multiple occasions in 2019 having had a series of falls, suffering a brain bleed and a broken pelvis, as well as a stint to be treated for a urinary tract infection. Mr Carter spent much of the coronavirus pandemic largely at his home in Georgia, and did not attend Joe Biden’s presidential inauguration in 2021, but extended his “best wishes”. Former first lady Rosalynn Carter, the closest adviser to Mr Carter during his term as US president, died in November 2023. She had been living with dementia and suffering many months of declining health. “Rosalynn was my equal partner in everything I ever accomplished,” Mr Carter said in a statement following her death. “She gave me wise guidance and encouragement when I needed it. As long as Rosalynn was in the world, I always knew somebody loved and supported me.”
